چکیده

Current-clamp experiments on an identified neurone have demonstrated the presence of L-glutamate receptors in the insect central nervous system. The cell body of the fast coxal depressor motor neurone (Df) in the metathoracic ganglion of the cockroach Periplaneta americana exhibits a hyperpolarizing response to L-glutamate, accompanied by an increase in membrane conductance. The response is dependent on both intracellular and extracellular chloride concentration, but is not affected by changes in potassium concentration. The hyperpolarization reverses at — 82mV (the equilibrium potential for chloride), is mimicked by the action of L-aspartate, blocked by the antagonists picrotoxin and y-D-glutamylglycine (y-DGG) at high concentrations (1-Oxl0~moll~), and is enhanced by L-amino phosphonobutyTate (L-APB). The response is insensitive to glutamate diethyl ester (GDEE), cis-2,3-piperazine dicarboxylic acid (cis-2,3PDA) and D-amino phosphonobutyrate (D-APB). The L-glutamate-activated increase in chloride conductance does not cross-desensitize with the y-aminobutyric acid (GAB A) response on the same cell. It is less sensitive than the GAB A response to block by picrotoxin. In addition, y-DGG specifically blocks the L-glutamate receptor.

A GABAB receptor on an identified insect motor neurone

The vertebrate GABAB receptor (GABABR) agonists 3-aminopropylphosphonous acid (3-APPA), SK&F97541 and 3-aminopropylphosphonic acid (3-APA) are able to induce a hyperpolarization on the cell body of motor neurone Df in the metathoracic ganglion of the cockroach (Periplaneta americana), although the classic vertebrate GABABR agonist l-baclofen fails to induce any responses on the same neurone. Voltage-independent Block of a Neuronal Nicottnic Acetylcholine Receptor by W-methyl Lycaconitine

TV-Methyl lycaconitine is the most effective low molecular weight antagonist reported for an insect neuronal nicotinic receptor. In the present study, the citrate salt of this neurotoxin from the plant Delphinium brownii was found to inhibit [H]-o^bungarotoxin binding to nerve cord extracts of the cockroach Periplaneta americana with a K, of l^x lO^ 'mol l" 1 . At a concentration of 1-Oxl0~moll...

SHORT COMMUNICATION A SULPHYDRYL REDUCING AGENT, DITfflOTHREITOL, MODIFIES AGONIST-NICOTINIC RECEPTOR INTERACTION IN AN IDENTIFIED INSECT NEURONE

Nicotinic acetycholine receptors are present at high density in the nervous system of insects (Sattelle, 1980). Studies on the structure of nicotinic acetylcholine receptors have shown a degree of homology between insect a-like subunits and a-subunits from other species (Bossy etal. 1988; Marshall etal. 1990; Sawruk etal. 1990a). The locust ar-subunit, when expressed in Xenopus oocytes, is able...

Electrical Characteristics of the Membrane of an Identified Insect Motor Neurone

1. The electrical properties of the membrane of an identified locust motor neurone, the fast extensor tibiae in the metathoracic ganglion, have been investigated to determine: the distribution of excitable and inexcitable membrane; the impulse initiation zone; and the conduction velocity of the spike in the ganglion and in the axon. 2. The waveform of extracellularly recorded spikes indicates t...

(S)- 3,5-Dihydroxyphenylglycine )an agonist for group I metabotropic glutamate receptors( induced synaptic potentiation at excitatory synapses on fast spiking GABAergic cells in visual cortex

Introduction: (S)- 3,5-Dihydroxyphenylglycine (DHPG) is an agonist for group I metabotropic glutamate receptors. DHPG-induced synaptic depression of excitatory synapses on hippocampal pyramidal neurons is well known model for synaptic plasticity studies. The aim of the present study was to examine the effects of DHPG superfusion on excitatory synapses on pyramidal and fast-spiking GABAergic cel...
